Intravenous general anaesthetics at least seem to work by enhancing the effects of GABA or blocking the effects of glutamate. The protein theory is currently considered more likely to explain the process of anaesthesia, because general anaesthetics affect synaptic transmission rather than axonal conduction. Both the release of transmitter and the response of the postsynaptic receptors are affected. General anaesthesia involves three main responses of the nervous system: loss of consciousness, loss of motor reflexes and loss of sensory reflexes. Because areas of the brain responsible for consciousness are the most sensitive it is possible to administer anaesthetics at concentrations that produce unconsciousness without seriously depressing the cardiovascular and respiratory centres or the heart muscle, but the margin of safety is narrow.

You could go into a very deep sleep where you do not breathe properly and have difficulty waking up.

Cannabis and other drugs may have their own side effects on your mental health, like anxiety or psychosis. Using heroin or methadone with diazepam will also increase their combined sedative effects. This could lead you to increase the dose of the stimulant to make up for it, which could be very dangerous. If you start or stop smoking while you are taking diazepam, you may have to change your dose.

This is because cigarette smoke affects the amount of diazepam in your body. You should tell your doctor if you smoke and how much, so that they can prescribe the correct dose for you.

A small percentage of patients do not respond to the drug. These individuals are insensitive or unresponsive to the drug. An equally small percentage responds in an exaggerated manner. These patients are described as intolerant.

The main disadvantage of this route of administration is that most drugs do not taste nice. Nasal administration is often intended to have a local effect, as in the use of nasal decongestants, although certain drugs are given this way to have a systemic effect.
